How might one use a radioactive isotope to determine whether the oxygen in CO2 comes from sugar or oxygen gas?
Feed the organism sugar in which the oxygen atoms have all been labelled with O-18. If the CO2 coming out has mostly O-18 instead of O-16, then it comes from sugar; otherwise, it comes from oxygen gas.
